Well if you are referring to a gutter or down spout typically the best thing to do is use a water hose. Take the house and push it as far into the pipe as possible. Then turn the water one. It should force and blockage loose.

If the drain pipes are exposed, sometimes they can be removed, cleared, and re-installed. For drain pipes that are internal, the best and sometimes only way, is to use some sort of clearing tool. Usually this means a drain snake, often attached to a drill. In extreme cases, you may have to hire a specialist with a video camera.
