Once you have determined you have missed your period, in the morning after you wake is the best time since the urine is not diluted by any fluids that you have had to drink. When the urine is not diluted the test is better able to pick up and trace of the hormone HCG.

If you are worried that you may be pregnant due to a missed period or pregnancy symptoms then you should go buy a pregnancy test from your nearest drug store. The best time to take an at home pregnancy test is first thing in the morning.

However, hormone levels can vary greatly at this point so a negative result doesn't necessarily guarantee you're not pregnant. If your period is already late, it's a good time to test.

Home pregnancy test can be take up to five days before your missed period but are most accurate the day of your missed period. It is best to take the test first thing in the morning because more of the pregnancy hormone will be present in your urine.
