No, though auditions are part of it. Casting directors representing the show contact agents they know when there is an opening on the cast. The agents then try to find someone in the stable of talent they represent that would be a good fit and then get him/her an audition.

Some of the players are chosen through auditions, but many are selected through recommendations or based on their previous experience. Kenan Thompson has been doing sketch comedy his entire career.
