Benadryl is a good treatment if the hotspots are allergic reactions, its a powerfull antihistamine. Use children's benadryl to be safe.

There could be a number of underlying issues causing the hot spots. Never medicate your animal when you don't know the cause you may make matters worse. Dogs generally don't have allergies so Benadryl likely won't work. Take your dog to your Veterinarian for diagnosis.

Benadryl can reduce itching in dogs and is safe to give one dose a day. Keep in mind your dog is itching from either fleas or allergies that are causing these hot spots, and it is recommended getting to the underlying problem before administering drugs that cover up the symptoms.

Benadryl only works in some dogs. If the itching has gotten to the "hot spot" stage, you may wish to see a veterinarian for a medication more effective for dogs.
