Moses was helped by the Holy Spirit at his mothers house when he was hidden from Egyptians. As an adult he first encounted the Holy Spirit at the Burning Bush.

In Exodus 3 we find Moses encountering the LORD God in the form of a burning bush. The location is Horeb, or "Mount Horeb". During this encounter (Exodus 4), God gives Moses miraculous signs, and Moses performs them. If you believe that Moses performed them with the help of the Holy Spirit, then this is the first occurrence.

The Holy Spirit descended upon Jesus's followers seven weeks after Easter, known as Pentecost. This is New Testament, not Old Testament- Moses didn't know the Holy Ghost, he was Jewish.
