This depends on what level you are coaching and your players. For high school kids, you want to run either a 4-4 or a 3-5. The benefits of having many linebackers is the prospect of blitzing. Most high school offensive lineman have trouble picking it up because of blocking schemes.

4-3, 3-4, 4-2-5, 4-4, and 3-3-5 are all widely used. All of those formations allowed for the linebackers (2nd number) to blitz and defensive backs to play zone.

Most defenses are based on a 3-4 or a 4-3 scheme. You're looking at having 3 or 4 defensive line men up front and/ or 3-4 line backers in the box. Your secondary will be have 2 CB, 1 SS, 1 FS. If you're coaching recreation or youth football, you do not need to work about too many schemes.

A lot of twists between the front defensive linemen, numerous blitzes, and a yoyo, when you drop the defensive linemen to the linebacker spot and the linebackers play the line.

The defense of today would have to be the three four scheme. The reason that it is used so much is because it defends the spread well which, is the most used offense in the world.

Playing either a 3-4 or 4-3 defense (essentially 3 defensive lineman/4 linebackers, or vice versa). Also, having four or five defensive backs (two cornerbacks, two safeties, etc. ). All of this is essentially based on the talent you have on defence. Once you evaluate that, you should then decide on what suits your team better.
