Video buffering just downloads the next few seconds of video before you actually see it. Thus, if your connection speed fluctuates, you'll still see the unbroken video. Making the buffer bigger would require you to wait longer to start watching the video, but might make playback smoother. For many applications, the buffer size cannot be changed.

If you have memory in your computer than yes you can make the buffer bigger if you know what you are doing, it will make videos stream a lot faster if you do this.
