first clean away loose dirt and dust use a wide paint brush starting at the top apply even strokes and work your way down the panel .There is a special spray gun you can buy from hardware stores which would be easier.

Apply the chosen color paint to the tongue and groove locations before mounting on the wall. This will eliminate a gap appearing without the proper color showing.

If it is old paneling, use a stuff broom to dust out the grooves. A stucco roller might get the paint in the grooves, but it could look lumpy. Using a brush, you can do the grooves first, then the whole panel. The best bet is a sprayer. Make sure to use a mask, however.
