When kittens are really young it can be very hard to tell if you don't know what you're looking for. Female kittens have more of "u" shape below their anus, whereas male kittens have more of an exclamation mark. When the kittens are a bit older it is easier to tell because the males' testicles will drop.

As with any other animal, you can check the kitten for male or female genitalia, which are generally very easily identified.
