Depending upon the desired complexity of the crossword puzzle, it is usually best to start with a grid of key words and then attempt to fill in as many additional words building off of them as possible. Have a dictionary handy to look up more obscure words that will fit into troublesome spaces.

Have a theme in mind whenever you start. Make a key first. Also, always try to write out where you want the words first rather than the boxes.
