Plotting your story very much depends on the type of story that you wish to tell. Usually, stories have 3 'acts'; the SET UP; which establishes your characters and setting, the INCITING INCIDENT, which sets out a problem or conflict your protagonists must overcome and the CONCLUSION, in which the protagonists achieve closure through resolving the aforementioned conflict.

You need an introduction to the characters and the situation. Then have the characters evolve to the desired resolution of the situation.

When coming up with a plot, always brainstorm first. Brainstorming can take many different forms and writing down every little conflict that comes to mind is a good way to decide on a plot. Be sure to avoid cliche's or the same type of conflicts used in other stories.

First you should decide on what the subject of your story is. Will this be a love story or a childrens' story? Then you need to decide on your characters.
