Do I respond to a rejection letter for employment?

Typically, there's no reason to respond to a rejection letter as it's generally their final decision. It depends on what the letter says, though. If they write that you'll be kept in consideration if future positions open up, or something like that, then you can send a brief letter of acknowledgment and thanks. But usually, rejection is rejection.

You don't have to. You can respond and say thanks for the consideration if you'd like but it won't really mean anything. It's better to say thanks after the interview.

I would say no why would you respond to a company that has rejected your employment this means they did not want you to work for them.
