Can you tell me how to take iron-on letters off?

Some iron-letters will peel right off, but most need to be reheated. I suggest warming an iron and placing it on the iron-on letters. Once the letters get warm, the glue gets soft and you can begin to tease apart the letters from the substrate. If the letters do not come off, try a higher heat setting.

Much the same way the went on, heat is your friend. Using your iron on high setting, get the iron-on letters very warm one at a time. After they are fully heated blast them with a good amount of steam from your iron. Repeat as neccesary until they are loose.

The most common way to get iron on letters off is to use an iron with the heat as high as possible, and if this leaves behind clumps of glue, spray hair spray onto them and then pick them off.
