I don't know if it's the largest, but the Winchester Mystery house is pretty big. 160 rooms with 40 bedrooms. It's a 4. 5 acre estate. Single houses aren't the thing anyway. Build a compound with a bunch of specialty buildings connected by tunnels. Bill Gates has a pretty large compound.

The largest house in the United States, to date is still the Biltmore Mansion. It has 250 rooms under one roof and is located near Ashevill, NC. There may be others with more square footage, but the Biltmore is considered the record holder.
