There is either a problem with the transmission or the linkage inside the transmission. It is also possible that you have a fuel leak that is causing your fluid to escape. Either way, you will need to replace the transmission.

check to see if the shift linkage is connected. also the gear indicator on the dash may be out of place.

The differential unit is seperate and requires gear oil, not automatic transmission fluid but that has nothing to do with it not going in reverse,if the fluid level is correct and the engine revs up but the car doesn't move in reverse then I would suspect a major transmission problem.
