How do I do a 2-spot color separation in Adobe?

In the print dialog window, click output in the options window. Click separations and "RIP". By the process names a spot color icon is displayed.

With Adobe Illustrator best thing is to know the number of your spot colors. You can assign them directly by clicking on the small arrow in the pantone palette.

First select file and then print. Select printer and then PPD file. Now you can select the type of mode that you want. The mode allows you to separate the colors as you desire. From here you can also scale or crop the image you are working with. For more instructions consult your users manual.
