Compressed refrigerant can be very dangerous and you can really mess up your ac system if your not careful. You should be able to refill the line on a specific port on the line. Make sure you fill the right line.

Look at the A/C lines on the passenger side of the engine compartment. Your fill connector will be located on the larger diameter, low side hose.

You should use a gauge to monitor the recharging process. Though not absolutely necessary,a gauge will help you recharge your A/C system more accurately, and reduce the chance of undercharging or overcharging. A high pressure A/C gauge can be connected tothe HIGH pressure service fitting, or alow pressure A/C gauge to the LOW pressure service fitting,or gauges can be attached to both fittings

Going to have to make some assumptions here since no vehicle information was provided. I will assume it is a later model car using R-134. Get a can of the R-134 refrigerant that has a charging hose and a gauge built into the can, connect it to the low pressure side of the system, follow the directions on the canister.
